    摘 要目的:探讨下丘脑雷帕霉素靶蛋白(mTOR)信号通路对雌性大鼠青春期启动的影响以及 mTOR 信号通路可 能的作用机制。 方法:选取清洁级雌性美国斯泼累格·多雷(SD)大鼠,随机分为 N 组(正常对照组)、二甲基亚砜(DMSO) 组(溶剂对照组)、雷帕霉素(Rapa)组。23 日龄时,对 DMSO 组及 Rapa 组大鼠行侧脑室置管术;29 ~ 35 日龄,Rapa 组大鼠予侧脑室注射 Rapa,DMSO 组予侧脑室注射等量 DMSO,N 组大鼠不予任何干预。每日对大鼠称重,28 日龄开始观 察阴门开启情况。35 日龄上午,末次侧脑室注射后 2 h 分别处死各组大鼠。分析各组大鼠体质量、阴门开启时间,子宫和 卵巢发育情况、血清黄体生成素(LH)、卵泡刺激素(FSH)、雌二醇(E2)水平、下丘脑 mTOR 信号通路相关基因和蛋白 表达量的变化。 结果:侧脑室注射Rapa后,与N组和DMSO组相比,Rapa组大鼠下丘脑p–mTOR及p–S6K1蛋白表达量下降, Rapa 组大鼠表现出阴门开启时间延迟,血清 LH、E2 水平显著下降,体质量、子宫湿重、卵巢湿重以及子宫指数、卵巢指数 显著降低,神经肽 Y(NPY)基因表达水平明显上升,差异均具有统计学意义(P < 0.05)。 结论:通过侧脑室注射 Rapa 抑制下丘脑 mTOR 信号通路,可延迟雌性大鼠青春期启动,其机制可能与 mTOR 信号通路影响下丘脑 NPY 表达水平有关。

    Abstract:

    AbstractObjective To investigate the effect of the hypothalamic mammalian target of rapamycin (mTOR) signaling pathway on puberty onset in female rats and the possible mechanism of the mTOR signaling pathway. Methods Clean grade female Sprague Dawley (SD) rats were selected and randomly divided into normal (N) group, dimethyl sulfoxide (DMSO) group, and rapamycin (Rapa) group. At the age of 23 days, lateral ventricular catheterization was performed on rats in the DMSO and Rapa groups. At the age of 29 to 35 days, rats in the Rapa group were injected with Rapa into the lateral ventricle, while rats in the DMSO group were injected with an equal amount of DMSO into the lateral ventricle. Rats in the N group received no intervention. The rats were weighed daily and the opening of the vulva was observed from the age of 28 days. On the morning of 35 days old, rats in each group were killed 2 hours after the last lateral ventricular injection. Changes in body mass, opening time of vaginal, development of uterine and ovarian, serum luteinizing hormone (LH), follicle stimulating hormone (FSH), estradiol (E2) levels, and hypothalamic mTOR signaling pathway related genes and protein expression in each group of rats were analyzed. Results After lateral ventricle injection of Rapa, compared with the N group and DMSO group, the expression levels of p-mTOR and p-S6K1 proteins in the hypothalamus in Rapa group rats decreased. The Rapa group rats showed delayed opening of the vaginal, and the serum levels of LH and E2, body mass, uterine wet weight, ovarian wet weight, uterine index, ovarian index and expression level of neuropeptide Y (NPY) gene were significantly higher in Rapa group rats (P < 0.05). Conclusion Inhibiting the mTOR signaling pathway in the hypothalamus through lateral ventricular injection of Rapa can delay the onset of puberty in female rats, and its mechanism may be related to the influence of the mTOR signaling pathway on the expression level of NPY in the hypothalamus.
