Abstract:〔Abstract〕 Objective To observe the effect and mechanism of Fuzheng Xiaoliu Granule on the migration of Her-2 positive breast cancer SKBR–3 cell line. Methods 5 groups of SKBR-3 cells were plated at the density of 2×106·2 mL-1 into 6 well culture plates and injured at 72h, 1 times dosage, 2 times dosage and 3 times dosage of Fuzhengxiaoliu granule containing serum were added in, Trastuzumab and blank serum were used as positive and blank control. After dosing 24h、48h、72h, Cell Scratch test were used to detect the cell migration of SKBR-3 cells; SKBR–3 cells were cultured for 48 hours, and the expression of Her–2 protein was detected by Western blot after 24 hours of treatment. Results The inhibitory effects of Fuzheng Xiaoliu Granule and Trastuzumab on the migration of SKBR-3 cells were signifi cant, Compared with the Blank control group, the difference were statistically signifi cant in 24h, 48h and 72h (P < 0.05~0.01); Western blot showed that 1 times dosage and 2 times dosage of Fuzheng Xiaoliu granule had no effect on the expression of Her-2 protein in SKBR-3 cells,compared with blank control group, there were no signifi cant difference (P > 0.05); The expression of Her-2 protein in the 3 times dose group of Fuzheng Xiaoliu Granule was signifi cantly lower than that in the blank control group (P < 0.05); Trastuzumab signifi cantly inhibited Her-2 protein expression, Compared with 1 times dosage, 2 times dosage of Fuzheng Xiaoliu granule and the Blank control group, the difference were statistically signifi cant(P < 0.01). Conclusion FuZheng XiaoLiu granule obviously inhibited the migration of breast cancer SKBR-3 cells; 3 times dose of Fuzheng Xiaoliu Granule and Trastuzumab inhibited the expression of Her-2 protein in SKBR-3 cells.
